The assessment of microbiological stability is an essential component of the overall stability programme for pharmaceutical products, as mandated by regulatory authorities worldwide including FDA, EMA, ICH, WHO, and national pharmacopoeias. While chemical and physical stability are routinely evaluated, the ability of a drug product to maintain its microbiological quality over its entire shelf life is equally critical to patient safety and product efficacy. Microbiological stability testing is not merely a routine quality control check; it is a systematic, risk‑based, and scientifically rigorous evaluation that demonstrates that the product remains free from objectionable microorganisms and that its preservative system (if present) continues to be effective under the intended storage conditions. A poorly designed or executed microbiological stability study can lead to undetected contamination, loss of preservative efficacy, and ultimately, product recalls or regulatory non‑compliance. Our ISO/IEC 17025‑accredited and GMP‑compliant laboratory offers a fully integrated microbiological stability testing service, covering the entire spectrum from protocol design, method validation, and sample testing to data interpretation, trend analysis, and regulatory submission support. We deliver quantitative, traceable results, including enumeration of total viable count (TVC), detection of specified and objectionable organisms, preservative efficacy (challenge testing), and, where applicable, Endotoxin/bacterial Endotoxin testing (BET). Our expert team of pharmaceutical microbiologists provides comprehensive study design, flawless execution, and clear reporting, enabling you to satisfy regulatory requirements, ensure product safety, and confidently establish your product’s shelf life.

Microbiological stability testing serves multiple critical purposes: (a) to demonstrate that the product meets its microbial quality specifications (e.g., total viable count, absence of specified pathogens) throughout its shelf life; (b) to confirm the effectiveness of any preservative system over the entire storage period, particularly in multidose products; (c) to identify any product‑ or package‑related microbial risks that may emerge over time (e.g., moisture ingress, package integrity failure); (d) to support the establishment of the product's shelf life and storage conditions; (e) to monitor batch‑to‑batch consistency and the robustness of the manufacturing process; and (f) to provide data for regulatory submissions (e.g., NDA, ANDA, MAA) and for responding to regulatory queries. The testing must be performed on multiple batches (typically three) and at multiple time points over the proposed shelf life, using validated methods and with appropriate controls. The choice of test methods – whether plate count, membrane filtration, or rapid microbiological methods (RMM) – must be justified based on the product type (sterile vs. non‑sterile), the dosage form (solid, liquid, semi‑solid), and the specific requirements of the pharmacopoeia (e.g., USP <61>, <62>, <71>, <51>, and EP 2.6.12, 2.6.13, 5.1.3). Our service begins with a comprehensive consultation to design a stability protocol that is scientifically sound and fully compliant with all relevant regulations.
We maintain a complete, validated suite of microbiological test methods, all performed in a dedicated pharmaceutical microbiology facility with stringent environmental controls, and using certified reference cultures (e.g., ATCC, NCTC) for method validation and quality assurance. Our portfolio includes:
2.1 total viable count (TVC) and Specific Microorganism Testing
We perform quantitative enumeration of total viable bacteria, yeasts, and moulds (TVC) using validated methods such as pour‑plate, spread‑plate, or membrane filtration, in accordance with USP <61> and EP 2.6.12. For non‑sterile products, we also conduct tests for specified microorganisms (E. coli, Salmonella, S. aureus, P. aeruginosa, C. albicans, and bile‑tolerant Gram‑negative bacteria) according to USP <62> and EP 2.6.13. We provide the results in CFU/g or CFU/mL, with full documentation of the method, the media, the incubation conditions, and the recovery validation. All tests are performed in duplicate or triplicate and include appropriate positive and negative controls.
2.2 Preservative Efficacy Testing (Challenge Testing) – USP <51>, EP 5.1.3
For multidose products containing antimicrobial preservatives, we perform preservative efficacy testing (PET) to demonstrate that the preservative system remains effective over the entire shelf life. We challenge the product at each stability time point with a panel of five test organisms (bacteria, yeasts, and moulds) and measure the log reduction at defined intervals (e.g., 7, 14, and 28 days). We provide the log reduction data, the statistical analysis, and a clear pass/fail determination according to the acceptance criteria of the relevant pharmacopoeia (USP <51> for categories 1–4, or EP 5.1.3 for criteria A and B). Our validated neutralisation procedures ensure that the recovery is not inhibited by the product’s preservative.
2.3 Sterility Testing – USP <71>, EP 2.6.1
For sterile products, we perform Sterility testing using the validated membrane filtration or direct inoculation methods, in accordance with USP <71> and EP 2.6.1. We test a representative number of units at each stability time point, under the appropriate incubation conditions (30‑35 °C for bacteria, 20‑25 °C for fungi), and we report a “pass” or “fail” determination. We also perform growth promotion testing on each media batch to confirm its suitability.
2.4 Bacterial Endotoxin Testing (BET) – USP <85>, EP 2.6.14
For parenteral products and devices, we perform bacterial Endotoxin testing (BET) using the gel‑clot, turbidimetric, or chromogenic kinetic methods, in compliance with USP <85> and EP 2.6.14. We provide the Endotoxin concentration in EU/mL or EU/device, and we confirm the absence of interference from the product at each stability time point.
2.5 Identification of Isolates – MALDI‑TOF, Sequencing, and Biochemical
If any colonies are detected during enumeration or Sterility testing, we perform definitive identification of the isolates using a combination of MALDI‑TOF MS, 16S/ITS sequencing, and biochemical profiling (e.g., VITEK, API). This allows us to distinguish between environmental contaminants, process‑related organisms, and actual product‑spoilage microorganisms, providing valuable data for the investigation of any out‑of‑specification (OOS) results.
2.6 Stability‑Indicating Method Validation – Full ICH Q2 and USP Compliance
All microbiological methods used in the stability programme are fully validated according to ICH Q2(R1) and the relevant pharmacopoeial chapters. We demonstrate specificity, linearity, accuracy, precision, limit of detection (LOD), limit of quantification (LOQ), and robustness for each method. The validation reports are provided as part of the overall stability documentation.
For clients requiring a deeper understanding of microbiological stability or for complex product types, we offer a suite of customised, research‑grade services that go beyond routine stability testing:
3.1 Accelerated and Stress Stability Studies – Predicting Long‑Term Performance
We design and execute accelerated stability studies (e.g., 40 °C/75% RH, 30 °C/65% RH) to predict the microbiological stability of the product under the worst‑case conditions. We also perform stress testing (e.g., freeze‑thaw, light, humidity) to assess the impact of extreme conditions on the microbial quality and preservative efficacy. The data are used to support the shelf‑life claim and to identify potential failure modes.
3.2 Container‑Closure Integrity Testing – Microbiological Challenge
We perform microbiological container‑closure integrity testing (CCIT) by challenging the sealed container with a suspension of Brevundimonas diminuta (or other relevant organisms) and detecting any ingress. This is essential for sterile products and for products that rely on the integrity of the package to maintain microbial quality.
3.3 Bioburden and Preservative Capacity Studies – Optimising Formulation
For products in development, we conduct Bioburden and preservative capacity studies to determine the natural microbial load and the ability of the formulation to control that load. This data is used to set appropriate specifications and to adjust the preservative concentration.
3.4 Data Analysis, Trend Reporting, and OOS Investigation
We provide comprehensive statistical analysis and trend reporting for the stability data, identifying any shifts in the microbial counts or preservative efficacy over time. In the event of an OOS result, we perform a thorough root‑cause investigation to distinguish between a genuine product failure and a laboratory error, and we provide a detailed investigation report.
3.5 Regulatory Submission Support – Preparation of Stability Reports
Our team of regulatory specialists assists you in preparing the microbiological stability sections of your regulatory submissions (e.g., CTD Module 3.2.P.8, or the stability summary). We provide a complete data package, including the method validation, the raw data, the statistical analyses, and a clear conclusion on the product's microbiological stability.
All analyses are conducted under a strict quality management system that includes the use of certified reference cultures, routine system suitability tests, and participation in proficiency testing schemes (e.g., the UK NEQAS for pharmaceutical microbiology). Our reports provide:
Our reports are structured to meet the requirements of regulatory audits and to facilitate a smooth review process. All data are stored in a secure, auditable environment, with full 21 CFR Part 11 compliance where required.
